| I. Background | |
|       信號轉(zhuǎn)導和轉(zhuǎn)錄激活蛋白(STAT)被確定為 DNA 結合蛋白的主要成分,該蛋白可響 應多種細胞因子而激活基因轉(zhuǎn)錄。STAT 家族中的七個不同成員(STAT1,STAT2,STAT3, STAT4,STAT5A,STAT5B 和 STAT6)調(diào)節(jié)從病原體到細胞因子分泌的許多生理和病理過 程。STAT4 被多種細胞因子激活,包括 IL12、I 型干擾素(IFN-1)、IL23、IL2、IL27、 IL35 等。 | |

|      STAT4-Luc HEK293 報告基因細胞,是由 STAT4 調(diào)控并表達 Luc 熒光素 酶報告基因的 HEK293 細胞。IFNγ與受體結合,激活 STAT4 的原理見下圖所示。
