                | I. Background | |
| cAMP 反應元件結(jié)合蛋白 (CREB) 位于細胞核內(nèi),是一種轉(zhuǎn)錄因子,在 Ser133 位點被不同的受體活化蛋白激酶(如蛋白激酶 A (PKA)、鈣調(diào)蛋白依賴性蛋白激酶 (CaMK)、絲裂原活化蛋白激酶 (MAPK) 和其他激酶磷酸化后,與靶基因啟動子的 cAMP 反應元件 (CRE)結(jié)合。CRE 是 CREB 的主要結(jié)合位點,負責激活 CREB。CRE 是許多細胞外和細胞內(nèi)信號通路的焦點,包括 cAMP、鈣、GPCR(G 蛋白偶聯(lián)受體)和神經(jīng)營養(yǎng)因子。cAMP/PKA 信號通路對許多生命過程和生物體至關(guān)重要。在 cAMP/PKA 信號通路中,CREB 通過 PKA 的磷酸化被激活,并與 CRE 結(jié)合,一般基序為 5'-TGACGTCA-3'。由于 CRE 是 cAMP/PKA信號通路的調(diào)節(jié)劑,因此可以研究各種抑制劑的作用。 | |
